多西环素抑制大肠癌细胞侵袭的实验研究

Effect of Doxycycline on invasion of human colorectal carcinoma LS174T cells in vitro

摘要: 目的观察多西环素(Doxycycline)对人大肠癌LS174T细胞体外侵袭的作用。方法 以人工重组基底膜(Matrigel)体外侵袭实验观察药物对细胞体外侵袭能力的影响;SDS-聚丙烯酰胺凝胶电泳法(明胶酶谱法)观察对金属蛋白酶分泌的影响。结果 多西环素抑制LS174T大肠癌细胞体外侵袭, 在多西环素浓度为5mg/L, 10mg/L, 20mg/L时, 每100倍视野穿过的细胞数分别为(101.3±6.7)个, (83.7±5.7)个和(42.0±2.0)个, 与对照组有显著差异, 抑制率分别为17.9%、32.0%和65.9%, 呈现剂量依赖性;在5mg/L的低浓度下多西环素即可抑制基质金属蛋白酶-2(MMP-2)的分泌。结论 多西环素能够抑制人大肠癌LS174T细胞的侵袭, 在低于抗菌浓度下即可抑制基质金属蛋白酶-2的分泌, 具有成为抗肿瘤侵袭药物的潜力。

Abstract: Objective To discover the inhibitory effect of Doxycycline on invasion of Human colorectal carcinoma LS174T cells in vitro.Methods Artificial recombinant basement membrane invasion assay was utilized through Transwell model.Zymography was used to observe the decrease of MMP2.Results Doxycycline could inhibit the invasion of human colorectal carcinoma LS174T cells in vitro.With 5mg/L, 10mg/L and 20mg/L Doxycycline, the number of cells which could invaded through the Matrigel was respectively 101.3±6.7, 83.7±5.7 and 42.0±2.0.The inhibition ratio was respectively 17.9%, 32.0% and 65.9%, dose-dependently.Under 5mg/L, Doxycycline could decrease the secretion of MMP-2.Conclusion Doxycycline had inhibitory effect on invasion of colorectal carcinoma LS174T cells through inhibition of the secretion MMP-2 under a low dose.Doxycycline was a potential agent inhibiting tumor invasion and metastasis.
