抗肿瘤药物白屈菜红碱的急性毒性试验研究

Study on the acute toxicity tests of the anti-tumor drug of Chelerythrine

摘要: 目的 测定动物一次给予抗肿瘤药物白屈菜红碱后的急性毒性反应和近似致死量。方法 应用小鼠、大鼠和犬等多种动物,按照药品审批规程要求进行一次给药后的观察。结果 动物出现安静少动,俯卧,呼吸深慢,精神萎靡,被毛疏松、无光泽,对弱刺激反应不敏感;死亡动物解剖发现胸腔有血性液体,多个脏器肿大、存在大面积充血和瘀血,尾静脉注射针孔处出血时间延长等异常变化。大鼠还出现明显的扭体反应和横向滚动等腹腔刺激症状。小鼠iv给药的LD50为 22.84mg/kg。大鼠ip给药的LD50为24.30mg/kg。Beagle犬iv给药的近似致死剂量为19.94mg/kg。结论 该药对动物具有明显的毒性反应,其致死原因与药液入血引起血管扩张,脏器多处充血或血液循环障碍,致呼吸障碍所致的急性呼吸衰竭和直接刺激腹腔各脏器有关。

Abstract: Objective To determine acute toxicity reactions and approximate lethal dose of the anti-tumor drug of Chelerythrine.Methods The drug toxicity and side effects were observed when the mice,rat and canine were treated according to toxicological procedures of safety evaluation for drugs.Results The animals were quiet and unanimated,breath was deep and slow,its fur was thin and lackluster;the rats were not sensitive to feeble stimulations.The dead animals were anatomized and abnormal changes were observed:liquid with blood cells in thorax,tumescence of many organs with congest and ecchymosis.A delayed bleeding time was observed in the pinhole of tail vein.Symptoms of celiac stimulation were also observed in rats,included the wring reaction and breadthwise welter.The median lethal dose(LD50)was 22.84mg/kg for mouse iv and 24.30mg/kg for rats ip.Approximate lethal dose was 19.94mg/kg for Beagle canine iv.Conclusions The drug on animals have significant toxicity,the cause of death related with the liquid into the blood caused by vascular dilation,congestion,multiple organ blood circulation disorders,acute respiratory failure caused by respiratory disorders and direct stimulation of abdominal organs.
