Effect of siRNA mediated silencing of cFLIP gene combined with Epirubicin on apoptosis of MCF-7 cells

Abstract: Objective To explore the effect SiRNA mediated silencing of cFLIP gene combined with Epirubicin on apoptosis of MCF-7 cells.Methods Chemosynthesis double strand siRNA of c-FLIP,transfer to MCF-7 cell,then detect the expression of c-FLIP and Caspase-8 protein using Western blot;detect the apoptosis with flow cytometry when c-FLIP-siRNA interference or combined with Epirubicin.Results c-FLIP-siRNA can inhibit c-FLIP protein expression effectively(P＜0.05),promote cells apoptosis(P＜0.05).Combined using of c-FLIP-siRNA and Epirubicin can promote MCF-7 cell apoptosis more significantly,compared with Epirubicin using alone(P＜0.05).Conclusion c-FLIP-siRNA can promote MCF-7 cell apoptosis,and increase the anti-tumor effect of Epirubicin.
