Bmi-1蛋白在结直肠癌组织中的表达及临床意义

Clinical significance of Bmi-1 protein expression in colorectal cancer and its precancerous lesion

摘要: 目的 分析Bmi-1与Ki-67,Bcl-2在结直肠癌中表达之间的关系,探讨其与结直肠癌临床病理因素的关系及临床意义。方法 采用免疫组织化学PV-9000法检测84例结直肠癌及配对正常肠黏膜,21例结直肠上皮内肿瘤中的Bmi-1、Ki-67和Bcl-2蛋白的表达。结果 Bmi-1在结直肠癌(73.81%)、上皮内肿瘤(95.23%)中的阳性表达率显著高于正常肠黏膜(59.52%),P<0.05。Bmi-1在结直肠癌中的表达与结直肠癌的WHO组织学分型、Dukes分期和淋巴结转移相关,P值均>0.05。在84例结直肠癌中Bmi-1的表达与Bcl-2的表达呈正相关,等级相关系数r=0.358,P<0.001。结论 结直肠癌和结直肠上皮内肿瘤中Bmi-1蛋白的异常表达可能与细胞增殖、凋亡和癌变有关。Bmi-1和Bcl-2在结直肠癌中的表达密切相关,表明Bmi-1可能协同Bcl-2发挥抗凋亡效应,但二者抑制凋亡途径不同。

Abstract: Objective To analyze the correlation between Bmi-1,Ki-67 and Bcl-2 expression in colorectal cancer and to explore the correlation and significance with clinicopathological features of colorectal cancer.Methods Immunohistochemistry staining was used to display the expression of Bmi-1,Ki-67 and Bcl-2 in 84 cases of colorectal cancer,84 cases of matched normal colorectal mucosa and 21 cases of precancerous lesion.Results The positive rates of Bmi-1 expression in colorectal cancer(73.81%)and intraepithelial neoplasia(95.23%)were significantly higher than those in normal colorectal mucosa membrane(59.52%),P<0.05.There was a positive relationship between Bmi-1 and Bcl-2 expression in 84 cases of colorectal cancer(r=0.358,P<0.001).Conclusion The abnormality of Bmi-1 expression in colorectal cancer might be involved in cell proliferation,apoptosis and cancerization.Bmi-1 was closely correlated with Bcl-2 in colorectal cancer,suggesting that Bmi-1 might coordinate with Bcl-2 to induce anti-apoptosis effect,whereas the suppressing apoptosis pathways were different.
