硫酸酯化茯苓多糖对胃腺癌细胞MKN-45凋亡的影响

Effects of sulfated derivatives polysaccharides from sclerotium of poria cocos on gastric adenocarcinoma MKN-45 cell apoptosis

摘要: 目的 探讨硫酸酯茯苓多糖(S-PCS3-Ⅱ)诱导胃腺癌细胞凋亡的作用。方法 体外培养低分化胃腺癌细胞株MKN-45,加入不同浓度的硫酸酯茯苓多糖作用于MKN-45细胞不同作用时间后,利用细胞形态学、流式细胞术(FCM)、DNA琼脂糖凝胶电泳、吖啶橙/溴化乙锭双染法、透射电镜等实验方法观察对细胞凋亡的影响。结果 硫酸酯茯苓多糖作用MKN45细胞后,细胞形态学观察见抑制细胞生长,浓度为0.5 g/L的硫酸酯茯苓多糖作用24 h、48 h、72 h后其凋亡率分别为(7.92±0.83)%、(20.68±1.75)%、(37.52±2.47)%,且DNA凝胶电泳出现梯状条带。荧光显微镜和电镜下可见典型的细胞凋亡形态变化。结论 硫酸酯茯苓多糖可诱导胃腺癌细胞凋亡来抑制胃腺癌细胞生长。

Abstract: Objective To investigate the effect of the sulfated derivatives polysaccharides from sclerotium of Poria cocos(S-PCS3-II)on gastric adenocarcinoma MKN-45 cells apoptosis.Methods The MKN-45 cell line was cultured in vitro,and was exposed to a range of concentrations of S-PCS3-II at different time.The effect of apoptosis was observed by cell morphology,flow cytometry,agarose gel DNA electrophoresis,fluorescent microscopy and transmission electronmicroscopy.Results S-PCS3-II can inhibit the growth of human gastric cancer MKN-45 cells.When MKN-45 cells were treated for 24h,48h,72h at 0.5g/L concentration,the apoptosis rate revealed(7.92±0.83)%、(20.68±1.75)%、(37.52±2.47)%,respectively.DNA agarose gel electrophoresis showed the typical DNA ladder of apoptosis.A typical apoptosis morphology was also observed by fluorescent microscopy.A typical apoptosis ultrastructure changes was also observed by transmission electronmicroscopy.Conclusion S-PCS3-II can inhibit the growth of human gastric cancer MKN-45cells by inducing gastric cancer cells apoptosis.
