芳香化酶抑制剂对胶原性关节炎大鼠血清中IL-17和RANKL/RANK/OPG的影响

Effects of aromatase inhibitors on IL-17 and RANKL/RANK/OPG in serum of collagen induced arthritis rats

摘要: 目的探讨第三代芳香化酶抑制剂阿那曲唑对胶原诱导的关节炎(CIA)大鼠血清中白介素17(IL-17)、核因子-κB受体活化因子(RANK)及其配体(RANKL)和骨保护素(OPG)的作用,了解其加重骨关节病变作用及其机制。方法 建立CIA大鼠模型,分为阿那曲唑组、卵巢切除组和假手术组,用排水法大鼠关节炎足体积变化,放射线分析大鼠关节病变情况,并用ELISA法分析CIA大鼠血清中IL-17、RANKL及RANK和OPG的表达情况。结果 与卵巢切除组比较,阿那曲唑组大鼠关节病变加重(P<0.05),血清中IL-17、RANKL表达增加,OPG的表达显著降低,RANKL/OPG比值明显增加,差异均具有统计学意义(P<0.05)。结论 在CIA大鼠模型中,阿那曲唑可增加大鼠血清中IL-17、RANKL的表达,而抑制OPG的表达,导致大鼠关节病变加重。

Abstract: Objective To explore the effect of aromatase inhibitors-anastrozole on IL-17 and RANKL/RANK/OPG in the serum of collagen induced arthritis(CIA)rats and to illustrate its aggravation on bone.Methods CIA rats were divided randomly into 3 groups:anastrozole group(A),ovarectomy group(O)and sham operation group(S).The edema rate of ankle joints were examined.The radiological injuries of ankle were observed by molybdenum target radiography.The production of IL-17 and RANKL/RANK/OPG in the serum were determined by enzyme linked immunosorbent assay(ELISA).Results Compared to ovarectomy group,administration of anastrozole significantly increased the severity of arthritis(P<0.05),the levels of IL-17 and RANKL in anastrozole group were significantly higher,whereas,OPG expression was lower.Moreover,the ratio of RANKL/OPG obviously increased(P<0.05).Conclusion Anastrozole potently promots the progression of CIA model by the increasing of IL-17、RANKL and decreasing of OPG.
