伏核微量注射异丁司特对吗啡成瘾大鼠甩尾痛阈的影响

Effect of ibudilast-microinjected into nucleus accumbens on appendix pain threshold in morphine-addicted rats

摘要: 目的 研究伏核(Nucleus accumbens NAC)内微量注入异丁司特对吗啡成瘾大鼠甩尾痛阈的影响。方法 正常大鼠及吗啡成瘾大鼠伏核经玻璃微电极注入异丁司特(5 μg/2 μL)或生理盐水(2 μL)后,以甩尾潜伏期(TFL)为指标,观察注药前后的镇痛作用。结果 正常大鼠及吗啡成瘾大鼠伏核内注入5 μg的异丁司特后,与注药前相比,其TFL延长,并且吗啡成瘾大鼠TFL延长较正常大鼠更为显著;Wistar正常大鼠及吗啡成瘾大鼠伏核注入2 μL生理盐水后,TFL无显著变化。结论 大鼠及吗啡成瘾大鼠伏核内微量注入异丁司特后,可使其产生镇痛作用,并且吗啡成瘾大鼠的镇痛效果更为显著。

Abstract: Objective The aim of this study was to investigate the effect of microinjection ibudilast on the appendix pain threshold in nucleus accumbens(NAC)of morphine-addicted rats.Methods Normal rats and morphine-addicted rats were injected with ibudilast(5 μg/2 μL)or saline(2 μL)into NAC through glass microelectrode.The tail-flick period(TFL)was used as an indicator to observe the thermal pain threshold before and after ibuilast injection.Results After injected ibudilast 5 μg into the NAC of normal rats and morphine-added rats,the TFL was prolonged compared with that before injection,and the TFL prolongation of morphine-addicted rats was more significant than that of normal rats.There was no significant change in TFL after 2 μL of saline was injected into the NAC of normal rats and morphine-addicted rats.Conclusion Microinjection of ibudilast into NAC of normal rats and morphine-added rats can cause analgesic effect,and the analgesic effect of morphine-addicted rats is more significant.
