A study of β-Glucuronidase in breast tissues and cancer by immune electron microscope

摘要:

目的 观察正常乳腺导管上皮细胞和乳腺浸润性导管癌细胞内β-葡萄糖醛酸酶在超微结构水平的定位及区别。方法 采用低温包埋、胶体金标记、免疫电镜技术,在透射电镜下观察正常乳腺导管上皮细胞和乳腺浸润性导管癌细胞内的β-葡萄糖醛酸酶。结果 标记β-葡萄糖醛酸酶的金颗粒存在于内质网和溶酶体内,同时观察到乳腺浸润性导管癌细胞中标记的β-葡萄糖醛酸酶金颗粒明显多于正常乳腺导管上皮细胞中的金颗粒数目。结论 本研究为进一步研究乳腺各种疾病和肿瘤中β-葡萄糖醛酸酶活性的变化,建立了实用的形态学模型。

Abstract:

Objective To identify the precise ultrastructural localization and difference of β-glucuronidase between the breast duct epithelial cells and the cells of invasive ductal carcinoma.Methods Low-temperature embedding,colloid gold labeling,and immunoelectron microscopy were performed,and the location of β-glucuronidase was observed under transmission electron microscope.Results The gold-particle-labeling β-glucuronidase was distributed in endoplasmic reticulum and lysosome.We found that the quantity of golden granules labeled β-glucuronidase in endoplasmic and lysosome of carcinoma cells was more than that in normal breast duct epithelial cells.Conclusion The morphological model of β-glucuronidase was established for further study in diseases,especially in neoplasms.
