Study on expression of mTOR、eIF4E and 4E-BP1 and their clinical significance in human cervical carcinoma

Objective To investigate the effects of the expression of mTOR、eIF4E and 4E-BP1 in the development and advancement of cervix carcinoma,in order to provide some valuable evidences for prognosis.Methods The expression of mTOR、eIF4E and 4E-BP1 in tumor tissues of 53 patients with cervix carcinoma and 15 cases with cervical intraepithelial neoplasia(CIN)and 13 cases with normal cervical epithelium were detected by S-P immunohistochemical technique.Results (1)The expression of mTOR、eIF4E and 4E-BP1 in tumor tissues were higher than those in CIN and normal cervical epithelial tissues(P<0.01),and there was no significant difference in mTOR、eIF4E and 4E-BP1 staining between normal cervical epithelium and CIN groups(P>0.05).(2)eIF4E positive-expression correlated significantly with disease stage、metastasis of pelvic lymph node(P<0.01),tumor cell differentiation and recurrence(P<0.05).(3)mTOR positive-expression was related to recurrence(P<0.01).(4)4E-BP1 positive-expression correlated significantly with disease stage(P<0.01).(5)mTOR positive-expression correlated significantly with eIF4E and 4E-BP1(P<0.01).(6)The analysis of single factor revealed that the expression of mTOR、eIF4E and 4E-BP1 was significantly related with prognosis of patients(P<0.01,P<0.05,P<0.05).Conclusion The increased expression of mTOR、eIF4E and 4E-BP1 may play an important role in the development and progression of the cervical carcinoma,which is associated with poor prognosis.

Objective To investigate telomerase activity of seborrheic keratosis and explore the relationship between telomerase activity and seborrheic keratosis.Methods Telomerase activity was detected using the modified telomeric repeated amplification protocol(TRAP-ELISA))assay in 30 seborrheic keratosis and 15 melanoma and 15 normal lesions obtained through open biopsy or resection.Results Telomerase activity of seborrheic keratosis was significantly higher than that in the controls,and this difference had statistical significance(P<0.05).Conclusion The findings suggest that telomerase activity should be associated with proliferation of seborrheic keratosis.
The efficacy of cryosurgery combined with chemotherapy in advanced non-small cell lung cancer

Objective To evaluate the feasibility and efficacy of using the Ar-He target cryoablation combined with chemotherapy for treating advanced non-small cell lung cancer(NSCLC).Methods Between 2005 Nov and 2009 Mar,90 non-small cell lung cancer patients confirmed by pathology were divided into the groups of chemotherapy after cryosurgery(group A,n=31),cryosurgery alone(group B,n=29)and chemotherapy alone(group C,n=30).The effective rate,the existence rate in one year and side effects were analyzed.Results The level of CEA decreased in the group A than that in the group B or group C.The response rate was 70.4%(19/27)in the group A,55.2%(16/29)in the group B and 42.9%(12/28)in the group C.There was a statistical difference between A and C group(P<0.05).The existence rate in one year was 63.0%(A:17/27),41.4%(B:12/29),46.4%(C:13/28),respectively,and there were no statistical difference between them(P>0.05).The side effects of all the three groups were tolerable.Conclusion The procedure using chemotherapy after cryosurgery have more efficacy than the cryosurgery alone or the chemotherapy alone,and the side effects are tolerable.The method is a valuable way for the advanced non-small cell lung cancer.

Objective To explore the association between smoking,alcohol consumption and p53 alteration in esophageal cancer among Chinese population.Method A systematic search was performed using the Medline and CNKI databases,and the eligible literatures were systematic reviewed.Results Fourteen studies were eligible for analysis,involving 2108 esophageal cancer cases and 1142(54.2%)p53 alteration cases.Among Chinese population,the pooled OR for the association between smoking,alcohol drinking and p53 alteration in esophageal cancer were 1.61(95% CI:1.09-2.38;P=0.003)and 1.91(95% CI:1.44-2.53;P<0.001),respectively.Conclusion The findings of this meta-analysis indicated that there were significant associations between smoking,alcohol consumption and p53 alteration in esophageal cancer among Chinese population.

Objective To investigate the therapeutic effect of natural killer cells(NK cells)on mice of primary liver cancer Heps transplant tumor and discuss the mechanism.Methods Mouse models of the solid tumor of liver cancer(Heps)were established.Then the mice were randomly divided into four groups:Normal saline(NS)group,treatment group 1(1×104 NK cells/mouse),treatment group 2(1×105NK cells/mouse),treatment group 3(1×106NK cells/mouse).Each group has 15 mice.At the same day,different concentrations of NK cells were injected into the mice through the tail vein.The NS control group was administrated with the same volume of NS.Measure the implanted tumors volume as well as the mean survival time was calculated.Two mice from each group were sacrificed at the 15th day respectively.Tumor tissue samples were collected for routine pathological section,hematoxylin-eosin(H-E)staining.The spleen NK cells activity was estimated by LDH release assay;the activated NK cells in spleen were studied by flow cytometer(FACS).Results At the 8th day after inoculating,the growth situation of the tumor of group 3 was slower than that of other groups(P<0.05).At the 20th day,the volume of the tumors was markedly decreased in the treatment group 1(P<0.05),and the treatment group 2 and treatment group 3 are even more decreased compared with the NS group(P<0.01).Treatment group 1 mice′ average survival is obviously longer than NS group and treatment group 2,3(P<0.01).Compare with the NS control group′s NK cell in splenic,the treatment groups are obviously higher(P<0.01).The lethality of the splenic was obviously enhanced in the treatment groups than that in the NS control group.Conclusions Moderate NK cells can effectively restrain the growth of Heps hepatocellular carcinoma transplantation,and extend the mice survival.It′s a experimental basis for the clinical application of NK cells.

Objective To study the effects of 2,3′,4,4′-tetramethoxybiphenyl(TMBP)on the anti-proliferation,apoptosis and expression of procaspase-3 in human cervical cancer HeLa cells in vitro.Methods HeLa cells were cultured in vitro and were incubated by TMBP in different concentrations.The cells were detected with MTT assay,fluorescent staining,DNA fragmentation assay,Western blot assay to observe the anti-proliferation and apoptosis-inducing effects,respectively.Results TMBP induced HeLa cells apoptosis in a time-and dose-dependent manner.Treated with 100M TMBP for 24h,nuclear morphological changes were observed by Hoechst 33258 staining.DNA fragmentation became obvious after various concentration TMBP-treated for 36h.Cleavages of procaspse-3 protein were observed with western blot assay.Conclusion TMBP could inhibited proliferation and induce apoptosis in HeLa cells,which may be related to the activation of caspase-3.

Objective To study spiral CT three-dimensional reconstruction technique for non-small cell lung cancer radiotherapy target volume delineation of the accuracy.Methods Application of spiral CT scan and TOMOCON image system.In lung lesions of normal two-dimensional CT and respectively by two-dimensional CT combined with spiral CT three-dimensional reconstruction technique target volume.The target volume is calculated and compared longitudinal diameter and size.Results 48 cases of lung cancer patients CT Tumor image:gross tumor volume(18.85~380.63cm3),median volume 103.60cm3;Two-dimensional CT combined with spiral CT three-dimensional reconstruction results:target volume(24.05~420.57cm3)median volume 130.55cm3.Two sets of data by matching t test,analysis results P= 0.037.Two groups of results showed that the average diameter difference is 1.5±0.5mm.Longitudinal diameter difference is 3.0±1.5mm on average.Conclusions Spiral CT three-dimensional reconstruction technique is advantaged to delineate non-small lung cancer radiotherapy target volume.It is a complement of conventional CT scan in non-small cell lung cancer radiotherapy target volume delineation.

Objective To investigate the relation between breast cancer and thyroid carcinoma.Methods Our study is a retrospective study including 8857 female patients in our hospital.Of these patients,650 patients suffered from breast cancer,the rest did not.There were 14 thyroid carcinoma in the group of breast cancer,42 thyroid carcinoma in the other group.Results The incidence of thyroid carcinoma was significantly higher in the group of breast cancer(2.15%)than that in the group without breast cancer(0.51%)(P<0.05).There were no apparently statistical difference in age,mean size of tumor,extracapsular invasion,nodal involvement and pathologic type between two groups.(P>0.05)Conclusions The patients with breast cancer have a higher incidence of thyroid carcinoma than the patients without.Ultrasonic and a fine needle aspiration by ultrasonic guidance should be routine examination.

Objective To study the clinical efficacy and economic effectiveness of biochemotherapy in the treatment of aging advanced non-small-cell lung cancer.Methods A total of 56 advanced non small cell lung carcinoma patients were random divided into two groups according to the different chemotherapy regimens applied by using cost-effectiveness analysis in pharmacoeconomics.Treatment group:Twenty-eight cases with non small cell lung carcinoma were enrolled and underwent chemotherapy with pemetrexed disodium,two cycle therapy followed by transfusion of CIK treatment,one cycle therapy.Control group:Twenty-eight cases with non small cell lung cancer were enrolled and only underwent chemotherapy,two cycle therapy.Results In treatment group,of the 28 cases,2 were CR,18 were PR,effective power is 67.9%;In control group,of the 28 cases,0 were CR,9 were PR,effective power is 32.1%.two groups contrast is obviously significant.Average cost is 37800 yuan in treatment group and 21038 yuan in control group,but cost-effectiveness in treastment group is lower than that in control group,two group contrast is obviously significant.Conclusions Patients with biochemotherapy can benefit from the combination of chemotherapy and autologous CIK transfusion.The treatment can enhance the efficacy of aging advanced non-small-cell lung cancer,and is a acceptable treatment modality in economic effectiveness.

Objective To discuss the clinical significance of intensity modulated radiation therapy(IMRT)in gerontal cervical cancer.Methods Sixty patients of gerontal cervical cancer in our study.Thirty patients were treated by IMRT,and thirty were treated by CRT.We compared short-term efficacy and complications between two groups.Results The tumour regression rates were 86.7% for IMRT group and 90.0% for CRT group(P>0.05)respectively.The 1-year and 2-year survival rates were 86.7% and 73.3% in IMRT group respectively,which were no significant difference compared with those in CRT group(80.0% and 63.3%),(P>0.05).Compared with CRT group,the doses and volume of small intestine,rectum,bladder in IMRT group were significantly decreased(P<0.05).The incidence rates of acute gastrointestinal tract and urinary system radiation reaction in IMRT group were significantly lower than those in the CRT group(P<0.05).The incidences of long-term radiation proctitis,cyctitis in IMRT group were significantly lower than those in the CRT group(P<0.05).Conclusion Compared with CRT in gerontal cervical cancer,IMRT did not improve local tumor control rate and the recent survival rate,but significantly reduced the response to acute and chronic radiation.

Objective To analyze the survival-related factors of breast cancer patients.Method SPSS 17.0 was used to analyze the correlation of tumor location,tumor size;postoperative clinical stage,pathological stage and ER status with 5-year survival of 174 breast cancer patients.Making Kaplan-Meier survival curves,comparative analysis.Result The age of high risk incidence of breast cancer was 40-49;The clinical stages is different,the survival curves are different.Tumor size and axillaries lymph node metastasis had positive correlation.The 5-year survival rate for postmenopausal patients was lower than that of pre-menopausal patients.ER(-)cases average disease-free survival was less than that of ER(+)cases.Conclusion Early detection,early diagnosis and early systemic therapy of breast cancer must be paid attention to.Breast cancer screening should be spread to the whole society.
