Expression and clinical significance of HABP-1 and VEGF in ovarian carcinoma

Objective To study the expression and clinical significance of HABP-1 and VEGF in ovarian carcinoma.Methods 20 cases with benign ovarian tumor,30 cases with border ovarian tumor and 102 cases with malignant ovarian tumor were used to measure the positive expression rate of HABP-1 and VEGF by immunohistochemical SP method.Results The positive expression rate of HABP-1 and VEGF was significantly higher in malignant and borderline groups than that in benign group(P<0.01).The positive expression rate of HABP-1 did not have association with age or histological type(P>0.05),but with the FIGO stage and metastasis(P<0.05).Conclusion The expression of HABP-1 had positive correlation with the expression of VEGF.Both of them are extremly useful and meaningful during the carcinogenesis,development and transformation.The expressions of HABP-1 and VEGF play an important role in diagnosis of epithelial ovarian cancer.

Objective To construct recombinant plasmids containing microRNA targeting the Pokemon gene and investigate the effects of miRNA-mediated downregulation of the Pokemon gene on the proliferation and cell-cycle progression of colorectal cancer cells,Lovo.Methods Four miRNAs were designed according to the coding sequence of the Pokemon gene and used to construct recombinant plasmids.The recombinant plasmids were transfected into Lovo cells using Lipofectamine 2000.The expression of Pokemon mRNA was detected by Quantitative polymerase chain reaction(qPCR)assays.Cellular proliferation was measured by methyl thiazolyl tetrazolium(MTT)assay.Cell-cycle progression was analyzed by flow cytometry.Results Four recombinant plasmids were successfully constructed,named mR22-1,mR22-2,mR22-3 and mR22-4.The expression of Pokemon mRNA was obviously downregulated in Lovo cells transfected with the recombinant plasmids.The best silencing effect was achieved in cells transfected with the mR22-1 plasmid.The expression levels of Pokemon mRNA were downregulated by 50%±2.7%.MTT assay indicated that mR22-1 transfection could inhibit cellular proliferation.Flow cytometry assay indicated that mR22-1 transfection lead to Lovo cell cycle arrest in G1-phase.Conclusion The recombinant plasmids containing miRNA targeting the Pokemon gene specifically downregulate Pokemon expression.Pokemon gene can promote proliferation in Lovo cells.Results suggest that the microRNA interference developed in this study represents a novel anti-tumor strategy that may be applicable to most research involving cancer therapy and,thus,has promising potential as a colorectal cancer treatment.

Objective To investigate AEG-1(Astrocyte Elerated Gene-1)and Ki-67 expression in different ovarian tissues and the relationship between them;to assess the clinical significance of AEG-1 expression in epithelial ovarian carcinoma(EOC).Methods Immunohistochemical staining was employed to detect the expressions of AEG-1 and Ki-67 in different ovarian tissues from 21 cases of normal tissues,32 cases of borderline tissues and 157 malignant tissues.The relationship between AEG-1 expression and clinicopathological factors was also assessed in these patients.Results We demonstrated that AEG-1 expression levels were gradually elevated in normal tissues,borderline tissues and EOC tissues,respectively.Moreover,there was a similar trend for Ki-67 expression(as a proliferative index)from normal tissues,borderline tissues to EOC tissues.Statistical analysis revealed that AEG-1 expression was markedly correlated with residue tumor size(P=0.0007),the FIGO stage(P=0.0063),Ki-67 expression(P<0.01)and histological differentiation(P<0.001)in the ovarian cancer patients.Conclusion AEG-1 overexpression in EOC may be associated with tumor progression,suggesting AEG-1 as a potential preventive target in these patients.

Objective To assess Hepsin expression in different ovarian tissues and in restigate the relationship between its expression and the clinical significance in epithelial ovarian cancer(EOC)patients.Methods Immunohistochemical staining was used to observe the expression of Hepsin in different ovarian tissues.The relationships of Hepsin expression,clinicopathological characteristics and responses to chemotherapy were also evaluated in ovarian cancer patients.Results We demonstrated that Hepsin expression levels were gradually elevated in normal tissues,borderline tissues and EOC tissues,respectively.Statistical analysis demonstrated that Hepsin expression status was positively correlated with the FIGO stage(P=0.0063),but negatively with histological differentiation(P<0.001)in EOC patients.Conclusion It is suggested that Hepsin over-expression in ovarian carcinoma may be correlated with its progression.

Objective To study killing effects of different dosage regimens of Gefitinib in combination with Cetuximab on Lovo cells(human colorectal cancer cell line)for exploring effects of Gefitinib and Cetuximab alone or combined on colorectal cancer.Methods Colorectal cancer cells were selected by direct sequencing of products after PCR amplification.Inhibiting effects of Gefitinib and Cetuximab of different times,concentrations and administration orders on Lovo cells with no K-ras mutation were evaluated by determination of combined effects of both drugs.Influence of optimal dosage regimen on cell cycle distribution and apoptosis rate of Lovo cells was determined by flow cytometry.Results Iit was confirmed that both Gefitinib and Cetuximab could induce apoptosis,and inhibitive effects of Gefitinib in combination with Cetuximab on Lovo cells were significantly stronger than that of Gefitinib or Cetuximab alone;However,different times,concentrations and administration orders of both agents did no produce significant difference(P>0.05).Conclusion Gefitinib in combination with Cetuximab could produce inhibitive effects on proliferation of Lovo cells.

Objective To estimate the appropriate margins for the planning target volume(PTV)in non-small cell lung cancer 3DCRT.Methods Eight non-small cell lung cancer patients under-went 3DCRTor IMRT.kV-CBCT scan was acquired on couch before and after radiotherapy.The analysis of these images was performed by automatic and manual regist ration of the kV-CBCT and planning CT images.The patient setup was then corrected by moving the couch accordingly.CBCT online-guided correction data were recorded.Results A total of 160 CBCT sets of 8 non-small cell lung cancer patients were analyzed.If the online image guidance was not corrected,the distance between the clinical target volume and the planning target volume was 10.9mm;If every online image guided radiotherapy was corrected,the distance between them was 2.2mm.Conclusion There is some extent of setup error in non-small cell lung cancer 3DCRT or IMRT.The approach based on the CBCT measurements can be used to reduce the impact of setup error obviously and to estimate the required margins for PTV.

Objective To study the expression of integrin-linked kinase(ILK) and E-cadherin in cervical intraepithelial neoplasia(CIN)and cervical squamous cell carcinoma,and analyze their relation with clinical pathological data.Methods Immunohistochemistry was used to detect the expression of ILK and E-cadherin in 54 cases of CIN,38 cases of cervical squamous cell carcinoma and 20 cases of normal cervical tissue.Results The positive expression rate of ILK in normal cervical,CINⅠ,CINⅡ~Ⅲ and cervical squamous cell carcinoma was 25.0%,68.8%,78.9% and 97.3%,respectively.The positive rate of ILK in CIN and cervical squamous cell carcinoma was significantly higher than that in normal cervix(P<0.05).The positive expression rate of E-cadherin in the normal cervix,CINⅠ,CINⅡ~Ⅲ and cervical squamous cell carcinoma was 100.0%,75.0%,55.3% and 36.8%,respectively.The positive rate of E-cadherin in CIN and cervical squamous cell carcinoma was significantly lower than that in normal cervix(P<0.05).There was a significant negative correlation of the expression of ILK and E-cadherin in cervical squamous cell carcinoma(r=-0.531,P<0.01).The positive expression of E-cadherin had correlation with clinical stage,differentiation and lymph node metastasis in cervical squamous cell carcinoma(P<0.05).Conclusion ILK and E-cadherin could be used as a marker to distinguish normal cervix from CIN and cervical squamous cell carcinoma.In addition,E-cadherin is also correlated with the malignant biological behavior of cervical cancer.

Objective To investigate the treatment pattern in patients with visceral organ metastatic breast cancer who having symptoms of brain or bone metastasis involved.Method The total dose of whole-brain radiotherapy were 45Gy and 40Gy,respectively.Ther bone radiotherapy were 33Gy and 40Gy,respectively.At the same time,chemotherapy were given with Paclitaxol 80mg/m2,d1、8、15,every 21 days as one cycle.The efficacy was evaluated every 2 cycles of chemotherapy according to WHO standard.Result The Objective efficacy of 4 patients were PR.The constricts symptoms of the CNS and pain of the bone were relieved.TTP of 4 patients were 9、5、6+ and 8.5 months,respectively. Conclusion Treatment pattern of concurrent chemoradiotherapy in visceral organ metastatic patients with having symptoms of brain or bone metastasis result in a effectiveness to reduce symptoms and improve QOL.

Objective To study the diagnosis and treatment of Gastrointestinal Stromal Tumors(GIST).Methods 32 cases of GIST which were admitted from January 2001 to December 2010 at the First People′s Hospital of Harbin were analyzed retrospectively.Results 32 cases of GIST were performed by laparotomy.5 of them underwent biopsy because of extensive metastasis.27 cases were resected completely.17 cases were followed-up.8 cases had recurrence and metastisis,and 3 of them were liver metastisis.Conclusion Surgery is the main therapy,and targeted therapy can improve survival.

Objective To compare the change of Ki-67,HER-2,ER and PR before and after neoadjuvant chemotherapy in breast cancer tumor,and explore the relationship between the changes and clinical response rate.Methods 103 patients with breast cancer were included in this study who didn′t accept any therapy such as radiotherapy,chemotherapy and endocrine therapy before recruiting.All cases were diagnosed as local infiltrating ductal carcinoma or lobular carcinoma by 14G hollow needle biopsy.94 patients were operated after 4 cycles chemotherapy.Tumor tissues were formalin-fixed,paraffin-embedded.Immunohistochemical staining was employed to detect the expression of Ki-67,HER-2,ER and PR.The clinical response rate was evaluated by WHO criteria.Results 9 patients of 103 cases had metastasis after neoadjuvant chemotherapy,and 94 cases underwent surgery after chemotherapy treatment.Pre- and postoperative tumor biopsy tissue sections stained were compared.The level of expression of Ki-67,ER and PR changed significantly,and HER-2 before and after the chemotherapy was not significant.However,only the change of Ki-67 expression was significantly correlated with tumor response rate.Conclusion Ki-67 expression as an indicator may be used to predict the tumor response rate during neoadjuvant chemotherapy of clinical outcome in breast cancer patients.
