Inhibiting vascular endothelial growth factor by RNA interference affects proliferation and angiogenesis of drug-resistant tongue cancer cell xenografts

Objective To investigate the effect of inhibiting vascular endothelial growth factor(VEGF)by RNA interference on proliferation and angiogenesis of drug-resistant tongue cancer cell xenografts.Methods Tca8113 cells were successively induced in vitro by cisplatin to acquire drug-resistance,and then injected subcutaneously in nude mice to establish xenograft models,which were randomly divided into non-transfected group,mock control group,scrambled sequence group and interference group.The transfection mediated by liposome was done in the latter 3 groups with intratumoral and peritumoral injection for inhibiting VEGF expression.Weight and size of xenografts were measured;immunohistochemical staining was applied to detect PCNA and CD34 for investigating tumor proliferation and microvessel parameters.In situ hybridization and Western blotting were used to test mRNA and protein of VEGF,respectively.Results The size,weight,mean optical density of PCNA protein and microvessel parameters of xenografts in interference group were less than those in other groups(P<0.05).The expression of VEGF mRNA and protein in interference group decreased significantly,compared by the other 3 groups(P<0.05).Conclusion RNA interference targeting VEGF could reduce proliferation and angiogenesis of drug-resistant tongue cancer cell xenografts through inhibiting VEGF expression,suggesting that inhibition of VEGF by RNA interference may serve as a new therapy for drug-resistant tongue cancer.

Objective To examine the anti-tumor efficacy and induction of apoptosis in combination therapy with low dose Rapamycin(RAPA)and Oxaliplatin(L-OHP)in HCT116 colon cancer cell line in vitro.Methods The growth inhibitory effect was evaluated by MTT assay in single agent or combination therapy.IC50 values were determined by using CalcuSyn 2.0 software.To determine interaction of the drugs,combination index(CI)were calculated by the method of Chou and Talalay.Apoptosis was investigated by flow cytometry and western blotting.Results Both RAPA and L-OHP inhibited cell proliferation in a does-dependent manner and the IC50 values were (8.35±0.78)μM (r=0.99)and (223.44±38.10)nM(r=0.94),respectively.The CI values were 0.52 and 0.72 when a combination of 10nM RAPA(IC20)with 1μM(IC20)or 2.5μM(IC30)L-OHP were used.The cytotoxicity of 10nM RAPA and 1μM L-OHP combination reached to 31%,which were nearly 2 folds of using L-OHP alone.The apoptotic rate of combination therapy after 24 hours treatment was higher than that of L-OHP alone,although RAPA at a subtoxic concentration of 10nm did not induce apoptosis.The cleaved-PARP protein expression level was the highest after 48 hours combination treatment.Conclusion RAPA at a subtoxic concentration in combination with low dose L-OHP synergistically inhibited HCT116 cancer cell growth in vitro.The mechanism was in relation to enhanced apoptosis.

Objective To observe the influence of SPS on the proliferation of PBMC and CD8+ cells in vitro.Methods PBMC extracted from healthy donors with ficoll-hypaque density gradient centrifugation were incubated with different doses of SPS in vitro.The proliferative activity of PBMC was tested by 3H-TdR assay and the proliferative activity of CD8+ cells was tested by fluorescence-activated cell sorter(FACS).Results SPS can accelerate the proliferation of PBMC with dose-dependent manner and was obvious at 1.25g·L-1 and 0.625g·L-1 dose group(P<0.05);SPS can obviously promote the proliferation of CD8+ cells(P<0.05).Conclusion SPS can promote the proliferation of PBMC and CD8+ cells,enhance the immune function of the organism.

Objective To evaluate the expression and clinical significance of A disintegrin and metalloproteinase(ADAM-19),Ki-67 in early invasive cervical cancer.Methods Expression of ADAM-19 and Ki-67 in 18 cases of normal cervical epithelium(NCE),22 cases of cervical intraepithelial neoplasm(CIN) and 82 cases of invasive carcinoma cervix(ICC)were detected by immunohistochemistry SP method.Results ADAM-19 was mainly expressed in the cellular membrane and/or cytoplasm in tumor cells,whereas the expression of Ki-67 was localized within the nuclei.It was demonstrated that positive rate of expression of ADAM-19 increased remarkably from normal cervical epithelium(NCE)to cervical intraepithelial neoplasm(CIN),and then to invasive carcinoma cervix(ICC)accordingly(P<0.05).In ICC,the expression of ADAM-19 correlated with pelvic lymph node metastasis,intravascular and stromal infiltration,FIGO staging,histological grading and Ki-67 expression(P<0.05);but not correlated with patients age and histological types(P>0.05).In cases with pelvic lymph node metastasis,intravascular and deeper stroma infiltration,FIGO staging II and histological grading III over-expression of Ki-67,positive rate of ADAM-19 was significantly higher than that in those without the conditions mentioned above(P<0.05).Conclution In cervical cancer,ADAM-19 may play an important role during proliferation of tumor cells,carcinoma invasion and metastasis.Over-expression of ADAM-19 may increase the proliferation of tumor cells and cancer progression,but it is not the only decisive factor.Early detection of ADAM-19 may be of value in further understanding the biological behavior and predicting the prognosis of cervical carcinoma.

Objective Expression of midkine (MK) in the laryngeal squamous cell carcinomas (LSCC) tissue were detected in order to probe into the relationship of the high expression of MK with the cervical lymph node metastasis,survival and prognosis of the LSCC.Methods All of samples were sectioned and were carried out by HE staining and MK immunohistochemical staining and MVD values were also detected.Results There was no or less expression of MK observed in 14 cases of the laryngeal normal mucous membrane and 14 cases of the laryngeal papilloma.There were 74.7% MK positive expression in 75 cases of LSCC,including 35 weak positive (+) cases and 21 strong positive (++) cases.MK is related with survival time(P=0.001),MK expression from negative to positive survival time decreased 5.63 times , Multivariate analysis showed that lymph node metastasis and recurrence related,The probability of recurrence with lymph node metastasis without lymph node metastasis was 5.488 times.Conclusion There were high expression of MK in the LSCC.The expression of MK was associated with the cervical lymph node metastasis.The expression of MK in LSCC might serve as an independent indicator for its diagnosis and prognosis.

Objective To evaluate the significance of diagnosis of isolation pulmonary nodules(SPN)using computer-aided diagnosis(CAD).Method 193 cases with SPN(including 144 cases of malignant nodules and 49 cases of benign nodules)proved pathologically by CT-guided biopsy in this paper,and analyzed the characteristics.According to the pathology results,Compare the accuracy of the neural network and artificial diagnosis of two methods in SPN's diagnostic.Result The diagnosis accurancy of neural network model was 71.5%,and the accurancy of artificial diagnosis was 80.3%,Sensitivity was 84.0% and 91.0%,respectively,specificity was 34.7% and 49.0% respectively.Conclusion Computer-aided diagnosis has better diagnostic accuracy rate,and the sensitivity of artificial neural network was closed to physicians diagnosis.It has certain significance in SPN diagnosis,can be a good diagnostic value,but with less accuracy.So it cannot completely replace the artificial diagnosis,but can be used as auxiliary diagnostic.

Objective To investigate the expression of protein fascin in normal thyroid tissues、thyroid benign lesions and thyroid papillary carcinoma and to evaluate their clinical significance.Methods The expression of protein fascin was determined in 60 patients with thyroid papillary carcinoma,40 of thyroid adenoma and nodular goiter and 20 of normal thyroid tissues by histochemical method(SP),then these values were compared among each other.Results The positive rate of fascin protein in thyroid papillary carcinoma(71.67%)was higher than that in thyroid benign lesions and normal thyroid tissues,there was significant difference between them(P<0.01).The positive rate in thyroid benign lesions was not higher than that in normal thyroid tissues,there was no significant difference(P>0.05).In thyroid papillary carcinoma with lymphatic metastasis,the positive expression of fascin protein significantly increased(91.30%).There was significant difference compared with no lymphatic metastasis(59.46%),(P<0.05).Conclusion The expression of fascin protein is correlated to the incidence,invasion and metastasis of thyroid papillary carcinoma and it may provide theoretical evidence for identification between benign diseases and thyroid carcinoma,it is also helpful for evaluating the behaviour of thyroid papillary carcinoma.The potiential of fascin protein as a novel early diagnostic marker and therapeutic target is assessed in thyroid carcinoma.

Objective To identify the precise ultrastructural localization and difference of β-glucuronidase between the breast duct epithelial cells and the cells of invasive ductal carcinoma.Methods Low-temperature embedding,colloid gold labeling,and immunoelectron microscopy were performed,and the location of β-glucuronidase was observed under transmission electron microscope.Results The gold-particle-labeling β-glucuronidase was distributed in endoplasmic reticulum and lysosome.We found that the quantity of golden granules labeled β-glucuronidase in endoplasmic and lysosome of carcinoma cells was more than that in normal breast duct epithelial cells.Conclusion The morphological model of β-glucuronidase was established for further study in diseases,especially in neoplasms.

Objective To observe expressions of FHIT and Livin in human non-small cell lung cancer(NSCLC)and investigate correlation and clinical significance between FHIT,Livin expression and infiltration,metastasis of NSCLC.Methods The expressions of FHIT and Livin protein in 60 cases of lung cancer and 19 cases of normal lung tissue were detected by SP immunohistochemistry method,and the clinicalpathology features of lung cancer were analyzed statistically.Results Compared with normal lung tissues,there was down-regulation of FHIT expression in NSCLC tissues(P<0.05),as well as up-regulation of Livin expression in NSCLC tissues(P<0.05).FHIT and Livin expression had close relationship with pathology type,cell differentiation degree,metastasis and TNM staging(P<0.05).FHIT and Livin expression had no relationship with patient' age,sex,tumor location and tumor size(P>0.05).Conclusion FHIT and Livin expression can be conducted as specific index to NSCLC prognosis.It will provide a new target to tumor gene therapy.

Objective To study the MRI manifestations and differential diagnosis of meningioma in lateral ventricle.Methods MRI findings,clinical and pathological data in 7 patients with meningioma within lateral ventricles were analyzed retrospectively.Results The main symptoms caused by meningiomas in lateral ventricles were headache and giddy。The tumors predominated in the trigone of left lateral ventricle,shown as roundish,lobulated soft mass,MRI findings of 6 benign cases were isointense on T1WI and iso/ slightly hyperintense on T2WI,markedly homogeneous enhancement,and the other 1 atypical meningioma presented heterogeneous signal intensity and heterogeneous enhancement.Conclusion Meningiomas in lateral ventricle have characteristic presentations on MR imaging,and when combined with clinical data,MRI can elevate the accuracy of diagnosis.

Objective To study the incidence,clinical features,diagnosis,treatment and prognosis of primary pulmonary lymphoma(PPL).Methods The clinical data of PPL patients treated in our hospital from January 1995 to January 2008 were retrospectively analyzed.Results 15 patients were diagnosed with PPL,accounted for 0.6% during the same period(15/2537).The advanced stage(stage Ⅲ and Ⅳ)patients accounted for 20%(3 cases).The 1-year overall survival and 5-year overall survival were 80.0% and 46.7% respectively.Surgical operation and chemotherapy were the major effective therapeutic approach for the treatment of PPL.Prognosis was favorable in patients with early stage of the tumor.Conclusion PPL is clinically rare and absence of typical clinicopathological characteristics,so its preoperatively exact diagnosis is difficult.Some specific imaging findings are helpful in the diagnosis of PPL.Final diagnosis of PPL could be made when a CT guided biopsy was done,followed by pathological examination with immunohistological study,or study of the tumor after surgery.The prognosis of patients received surgical operation is relatively better.
