The effects of ILK-ASODN on inhibition of formation of subcutaneous xenografts of human epithelial ovarian cancer in nude mice

Abstract: Objective To study the effect of Integrin-linked kinase antisense oligonucleotides(ILK-ASODN)transfected to human ovarian cancer HO-8910 cell lines on inhibition of xenografts of human epithelial ovarian cancer in nude mice.Methods The study was divided into control and experiment groups.We measured ILK mRNA and protein expression in the cells by RT-PCR and Western blot at 72 hours after transfection;evaluated the influence of ILK-ASODN transfection on the cell cycle by flow cytometry.After transfection,HO-8910 cells were subcutaneously inoculated into nude mice.We observed the changes of nude mice weight,in vivo tumor formation rates,the time for the tumor development,tumor volume and weight,and calculated the rates of tumor inhibition of the two groups.Results Compared with the control group,ILK mRNA and protein in the experimental groups were significantly lower(P<0.01);the number of the cells in G0/G1 phase were significantly lower(P<0.01),tumors in the nude mice of the experimental group developed highly significantly later(P<0.01).The speed of the tumor volume and weight growth were significantly slower in the experimental group than those in the control group(P<0.01).Conclusion ASODN blocked the ILK gene expression of human ovarian cancer cells and inhibited the growth of the cells;ILK gene silencing significantly inhibited the formation of the subcutaneous xenografts of human epithelial ovarian cancer in nude mice.
