The influences of Decorin gene on biological behavior in human lung adenocarcinoma A549 cell line

Objective To explore the influences of decorin(DCN)gene down-regulating on biological behavior in human lung adenocarcinoma A549 cell line.Methods Chemically synthesis in vitro targeting DCN-siRNA was transfected into human lung adenocarcinoma A549 cells by LipofectamineTM 2000.The gene expression of DCN was detected using Real-Time PCR;The protein expression of DCN was investigated using Western blot;Checkout DCN-siRNA effects on lung adenocarcinoma cancer cell proliferation was detected by CCK-8;The migration and invasion ability were determined by Transwell assay.Results DCN-siRNA was successfully transfected into human lung adenocarcinoma A549 cells.Real-Time PCR results showed that DCN mRNA expression level significantly decreased,compared with untransfected group,negative control group.Western blot results showed that DCN-siRNA transfection inhibited DCN protein expression level;CCK-8 results showed that the proliferation was enhanced in the DCN-siRNA group as compared with untransfected group,as well as the negative control group and empty vector group.Transwell assay results showed the invasion of A549 cells in DCN-siRNA group was significantly enhanced as compared with untransfected group[(22.6±1.14)vs.(5.2±0.84)].Wound-Healing assay results revealed that the cell repairing rate was markedly increased in DCN-siRNA group.A549 cells were close to complete repair after 48 hours.Apoptosis was not significant in DCN-siRNA group as compared with untransfected group(P=0.214).Conclusion Down-regulation the expression of DCN gene by DCN-siRNA may enhance the ability of invasion,migration and proliferation of A549 cells without affecting apoptosis,which provides a new evidence of function for advancing research of lung cancer.

Objective To explore the relationship between the types of female breast density and age and breast cancer.Methods By accepting the digital mammography X-ray examination for 5006 women cases and according to the ACR BI-RADS standard in the fourth edition,the breast density assessment was quantified.We analysed the relationship between the breast density and age and breast cancer.Results In 5006 cases,the average female age was between 44.22±8.09 years old,median age was 43 years old.The components of the breast density were fat type,small amount type,large amount type and compact type each count were 256(5.11%),726(14.51%),3719(74.29%),305(6.09%)respectively.By dividing into different age-group to analyze the breast density,there was significant statistical differences of the breast density among age-groups(P<0.001).Among them the breast cancer were 184 cases.Age was between 51.26±10.15 years old.Breast cancer in each breast density were fat type 10.16%(26/256),small amount type 9.09%(66/726),large amount type 2.45%(91/3719)and compact type 0.33%(1/305).There were statistical differences among age-groups and breast densities and breast(P<0.001).Conclusion Age plays a very important effects on the female breast density.The lower breast density is a high risk factor to breast cancer occurrence.

Objective To investigate the efficacy and safety of HCPT combined with 5-fluorouracil(5-Fu)in treatment of bladder cancer.Methods Selected 92 patients with advanced bladder cancer in our hospital from February 2011 to August 2015,equally divided into observation group and control group of 46 patients based on the random number table,the control group were received 5-Fu intravesical chemotherapy,The observation group were given the HCPT combined with 5-Fu chemotherapy,the chemotherapy infusion were once a week,continuous perfusion were observed for 3 months.Results The response rates in the observation group was 95.7% and the control group was 76.1%,the observation group were significantly higher than the control group(P<0.05).The occurrence of bladder irritation adverse reaction,blood abnormalities,liver and kidney dysfunction,bone marrow suppression in the observation group during chemotherapy compared with the control group were no significant difference(P>0.05),all adverse reactions after symptomatic treatment were improved.All patients were followed up for 1 year,The mortality and recurrence rates in the observation group were 8.7% and 4.3% respectively,while the control group were 30.4% and 17.4%,the mortality and recurrence rates of the observation group were significantly lower than the control group(P<0.05).Conclusion HCPT combined with 5-Fu chemotherapy for bladder cancer has strong anti-cancer effect and without increase the incidence of adverse reactions,it can promote the improvement of the long-term prognosis,so it is a safe and effective topical bladder chemotherapy drugs.

Objective To analyze cancer incidence and mortality in Fujian in 2012 and to provide scientific basis for tumor prevention.Methods In accordance with the Methods and criteria of data quality control made by NCCR,7 regristries data qualified from 9 submitted regritries in Fujian after data assessment were merged and analyzed.Results In 2012,the cancer incidence rate was 251.44/105(308.44/105 in male and 193.03/105 in female),age standardized incidence by Chinese standard population(ASR China)and by world standard population(ASR world)were 205.15/105 and 201.11/105.The cumulative incidence(0~74 age)was 23.51%.The mortality rate was 161.85/105(220.87/105 in male and 101.37/105 in female).ASR China and ASR world were 128.54/105 and 127.70/105 the cumulative incidence(0~74 age)was 15.04%.The age-specific incidence and mortality reached maximum value in 75 ages and 80 ages respectively.The top 5 cancer incidences were lung cancer,stomach cancer,liver cancer,esophagus cancer and breast cancer.The top 5 cancer mortality were lung cancer,liver cancer,stomach cancer,esophagus cancer and colorectum cancer.Conclusion Digestive malignancies,lung cancer,and breast cancer in female were the most frequent in Fujian province,and the prevention and control for those cancers should be enhanced.

Objective The study is mainly on the effect and outcome of 131I treatment for children and adolescents with differentiated thyroid cancer.Methods We compared the difference between rhTSH-aided radioiodine treatment and L-thyroxin withdrawal.Our study collected 87 patients in a retrospective study.In order to analysis the effect,we divided into two groups.The experimental group has 46 patients,and control check has 41 patients.We compared the basic characteristics and mortality in different groups by χ2-test,t-test and F-test based on data type.Results The thyroid stimulating hormone concentrations of experimental group patients have significant differences in the first,the third and the sixth day after the injected of rhTSH,the third day is the highest.The TSH concentration has significant difference between the two groups before 131I treatment(t=2.362,P=0.023).For the Tg(thyrobolulin)negative patients,Tg concentration in control check is higher than the third day after injected of rhTSH in the experiment group(1.5±1.2 vs. 0.7±1.4,P=0.034).Whole body image has been done by scintigraphic after 3~8 months of 131I treatment.Thirty four(84%)patients have no radioiodine uptake in experiment group,and 40(87%)patients have no radioiodine uptake in control check,there is no difference between the two groups(χ2=0.277,P=0.599).There is no difference between the two groups on the reason for retreatment(P=0.875).Conclusion The outcome of rhTSH-aided radioiodine treatment is similar to L-thyroxin withdrawal in short-term recurrences,and rhTSH-aided radioiodine treatment with 131I therapy for children and adolescents with DTC is a better method.

Objective To explore the intraoperative process and determine that the stomach resection margin is accurate so as to ensure the quality of operation and improve the patients′ survival rate.Method Normally,tissue samples were continuously collected from two sides of visualized tumor edge as center.Specifically,the tissue samples were collected from upper side when tumor located at gastric antrum,collected from lower side when tumor located at gastric cardia,and collected from distant side when tumor located at gastric body.All samples were processed with paraffin embedding,staining,and were judged by the distance of tumor cell infiltration.Results The mucosa infiltration distance was more than 5 cm in 5 cases,more than 3 cm but less than 5 cm in 5 cases,and less than 3 cm in 118 cases.A total of 218 cases had less 0 cm mucosa infiltration distance,which was visualized distance less than the microscopic.The serosa infiltration distance was more than 5 cm in 6 cases,more than 3 cm but less than 5cm in 19 cases,and more or equal than 0 cm in 251 cases.A total of 95 cases had less 0 cm infiltration distance.The results of the single factor analysis showed that whether the general observed infiltration distance of the mucosal membrane was greater than what observed under the microscope,is associated with gender,age,maximum diameter,thickness of the tumor,external quality,and TNM stage.It significantly demonstrated the relationship of microscopic mucous membrane infiltration distance with sex,vertical diameter,thickness,stroma mass,and TNM stage.Stroma membrane infiltration distance significantly correlated with vertical diameter,the largest diameter,thickness,gross type,histological type,stroma mass,and TNM stage.Conclusion The infiltration of distance was less than 5 cm among ninety nine percent of patients with gastric cancer.Therefore,enlarged resection of 5 cm from visualized margin of tumor is necessary for operative treatment.There are associations between tumor infiltration distance and vertical diameter,thickness,stroma mass,manner of infiltration,and TNM stage.

Objective To explore the clinical characteristics and the influence factors of prognosis of advanced ovarian cancer patients after abdominal metastases.Methods We retrospectively analyzed 65 cases during January 2013 to January 2016 in the First People′s Hospital of Tianmen.The patients were diagnosed clearly with pathological,diagnosed for the first time and has the complete clinical data including peritoneal metastases in 58 cases,which were analyzed for single factor and multiple factors of influencing factors survival analysis.Results 58 cases of patients with diagnosis of peritoneal metastasis in ovarian cancer that the average is(49.2±6.5),from ovarian cancer diagnosis to abdominal metastases for an average time of 11 months,ovarian cancer patients with peritoneal metastasis of the median survival time was 8 weeks,but 7 cases without abdominal metastases that the median survival time was 15 weeks.The single factor survival curves(Kaplan-meier)showed that marital status,reproductive history,history of breastfeeding,malignant ascites,neoadjuvant chemotherapy,comprehensive treatment(chemotherapy and tumor cells to destroy the loss and laparotomy abdominal hot perfusion chemotherapy),peritoneal metastasis tumor number,residual lesion size,quality of life in patients with residual lesions(KPS scores),D-dimer level in plasma and urine trace albumin levels related to the prognosis of patients(P<0.05).Conclusion Patients with advanced ovarian cancer patients with peritoneal metastasis survival time is shorter,and also company with poor prognosis;Neoadjuvant chemotherapy combined tumor cells to destroy the loss and postoperative intraperitoneal hot perfusion chemotherapy,short-term curative effect is good,it can not only improve the advanced ovarian cancer treatment effectiveness,but also improve the quality of life in patients of late phase.The D-dimer level in plasma and urine trace albumin levels before treatment have remained in the patients with high level or not reduce,treatment effect and prognosis was poor.These can be used as a new index of judging prognosis.

Objective To evaluate the clinical effect of Uniportal video-assisted thoracoscopic surgery and single utility port video-assisted thoracoscopic surgery for ⅠA stage non-small cell lung cancer.Methods A total of 81 patients with ⅠA stage non-small cell lung cancer was admitted to our hospital from January 2015 to November 2015.The patients were divided into control group and observation group.51 patients of control group received single utility port video-assisted thoracoscopic surgery,while other 30 patients of observation group received Uniportal video-assisted thoracoscopic surgery.Results The incision length of observation group was(4.27±0.29)cm,operative time was(208.80±61.12)mins.The volume of blood intra-operation was(92.33±73.75)mL;the number of lymph nodes dissection was(15.62±5.12),post-operative drainage within 24 hours was(401.70±53.31)mL;drainage tube retention was(6.30±3.01)days;postoperative hospitalization time was(19.03±5.85)days.The incidence rate of postoperative complications was 13.33%.The incision length of control group was(3.86±0.23)cm.The operative time was(184.30±51.36)mins;The volume of blood intra-operation was(84.90±80.98)mL,the number of lymph nodes dissection was(15.84±5.66),post-operative drainage within 24 hours was(398.00±52.73)mL;drainage tube retention was(6.10±3.25)days;postoperative hospitalization time was(18.69±6.81)days;The incidence rate of postoperative complications was 25.49%,there were no significant difference between the two groups(P>0.05).VAS pain score of the observation group was(2.32±0.94)and it was(4.18±0.95)in control group,and observation group was significantly better than the control group(P<0.05).Conclusion Uniportal video-assisted thoracoscopic surgery has the benefit of less trauma and pain,and it is worth using widely.

Objective To study the effects of anti-cancer drug NSC319726 on the gene expression of ovarian cancer.Methods The data that NSC319726 acted on the ovarian cancer cell lines TOV112D was downloaded from the GEO database and then the bioinformatical analysis was conducted.Results NSC319726 increased 246 genes including MMP3,CXCR4 et al and decreased 198 genes such as PBX1 in ovarian cancer cell lines.GO analysis indicated that six GO items were related to metabolism.Pathway analysis revealed that NSC319726 could affect the circadian clock and MMP signaling pathway.Conclusion NSC319726 could affect the expression of multiple functional genes in ovarian cancer cell lines and might be a potential drug targeting to the precursor-B cell acute lymphoblastic leukemia.Meanwhile,it should be noted that NSC319726 may have effects on the tumor metastasis and human sleep.Therefore,further research needs to be conducted before the clinical use.

Objective To analyze the relationship between Arg-1 expression and the clinical pathological factors,proliferation and prognosis value in patients with colorectal cancer.Methods The expression of Arg-1 was observed in normal tissues,chronic inflammatory tissues,and adenomas inflammatory carcinoma tissues of mice.At the same time,Arg-1 expression was observed in human colorectal cancer adjacent tissues,inflamed tissues and colorectal cancer tissues.Arg-1 expressed in 20 cases colorectal inflammation-cancer model in mice.Arg-1 expressed in 20 normal colorectal tissues.Fiftheen colitis tissues and 110 colorectal cancer tissues were examined by Immunohistochemistry.Statistical analysis was used to analyze the changes of Arg-1 expression in different groups of mice and human colon tissue cases.Results Arg-1 protein expression in normal tissues of mice was gradually increased in colon,chronic inflammatory tissues,adenomas,inflammatory carcinoma,with statistical significances(P<0.05).Arg-1 expression in para-carcinoma tissue,colitis tissues,colorectal cancer tissues was gradually increased with statistical significance(P<0.05).Conclusion Arg-1 protein is associated with colorectal cancer TNM stage.Arg-1 protein may be involved in occurrence and development process of inflammation-associated colon tumor and may be a candidate of proliferated and prognostic biomarker in patients with colorectal cancer.

Objective To discuss the effect of microRNA-27a on U251 glioma cells.Methods Overexpression or inhibition of miR-27a in U251 glioma cells were done by transient transfection of miR-27a mimics or AMO-27a in vitro.Cell viability was detected by MTT assay.Invasion ability of U251 was detected by transwell invasion assay.The level of miR-27a and PPARγ were detected by real-time PCR.Results Under inhibition of miR-27a condition,the proliferation and invasiveness of U251 glioma cells were decreased.The level of PPARγ was significantly increased,whereas the level of miR-27a was decreased.Overexpression of miR-27a increased the proliferation and invasiveness of U251 glioma cells and decreased the level of PPARγ.Conclusion Inhibition of miR-27a is benefit for inhibiting the proliferation and invasiveness of U251 glioma cells.

Objective To investigate the clinicopathological significance of upregulated NQO1 protein expression in uterine cervix carcinoma(UCC).Methods Immunohistochemical staining was performed on paraffin-embedded UCC specimens from 123 patients.Disease-free survival(DFS)and overall survival(OS)rates for all cervical UCC patients were calculated using the Kaplan-Meier method,and univariate or multivariate analyses were performed using the Cox proportional hazards regression model.Results The NQO1 protein showed a mainly cytoplasmic staining pattern in cervical cancer cells,and the strongly positive rate of NQO1 was significantly higher in UCC.High-level NQO1 was closely associated with poor differentiation,late-stage,lymph node metastasis and high-risk for HPV infection.Additionally,high-level NQO1 was associated with lower DFS and OS rates.Furthermore,Cox analysis revealed that NQO1 expression emerged as a significant independent hazard factor for DFS rate in patients with UCC.Conclusion NQO1 overexpression might be an independent biomarker for prognostic evaluation of UCCs.

Objective To evaluate the clinical efficacy and safety of celiac continued circulatory hyperthermia perfusion with chemotherapy in the treatment of cancer with ascites.Methods Databases including the Cochrane Library MEDLINE,PubMed,Embase,CBM,VIP,CNKI and WanFang Data,and the other sources as supplying.The literatures were screened according to the inclusion criteria,extracted data and assessed the methodological quality,and then meta-analysis was performed using RevMan 5.0.2 software.Results Totally 14 RCTs with 867 patients were included.Meta-analysis showed that compared intraperitoneal chemotherapy,with celiac continued circulatory hyperthermia perfusion had a significant difference in CR(OR=3.18,95%CI:2.18~4.64,P<0.00001),PR(OR=2.05,95%CI:1.51~2.77,P<0.00001),overall effective rate(OR=4.88,95%CI:3.59~6.46,P<0.00001).The incidence of adverse reactions was no statistically significant in the two groups.Conclusion Celiac continued circulatory hyperthermia perfusion with chemotherapy in the treatment of cancer with ascites demonstrates better efficacy and safety,but it still needs to verify the above conclusion.

Recent studies had demonstrated that Renin-angiotensin system(RAS)plays an important role not only in the cardiovascular system,but also in the occurrence and development of breast cancer.Associated with RAS receptor,Angiotensin Ⅱ(Ang Ⅱ)and Angiotensin 1-7(Ang1-7)which are the major molecules of the RAS,respectively,promote or inhibit the occurrence and development of tumor.In this review,we summarize the current knowledge of the RAS in breast cancer,and discuss the potential application of RAS related receptors targeted drugs in the clinical treatment of breast cancer.

Metastasis is the leading cause of death in patients with lung cancer.Circulating tumor cells(CTCs)play a key role in the process of distant metastasis.In recent years,research results have shown that CTCs may become a emerging marker and new target in the treatment of lung cancer.In this article,we elaborate the clinical significance of CTCs in lung cancer from the way of diagnosis and clinical staging,metastasis and recurrence,individual treatment and prognosis and so on,so that we can provide a new direction for the treatment of lung cancer.

Pancreatic adenocarcinoma(PAC)is still a refractory human digest malignancies due to multifaceted causes,late diagnosis and insensitive to traditional chemo-and radio-therapy.Resistance to apoptosis could be one of the most relevant mechanisms for PAC to escape any non-surgical therapy.This review aims to clear up the main deregulated apoptosis signal pathways over the years and to find out the abnormal molecule target(s),and therefore,provide novel concepts for PAC molecular targeting therapy.

MiRNA is a class of small endogenous non-coding molecule RNA with regulatory functions,which regulates the target genes at the post-transcriptional level by binding with 3′untranslated regions(3′UTR)specifically.Nuclear factor kappaB(NF-κB)is a kind of significant intranuclear transcription factor.Being activized,it easily combines with the fixed nucleotide sequence in promoter region to initiate gene transcription and participates in the invasion and metastasis of gastric cancer.Recent researches showed that miRNA not only influences on tumor invasion and metastasis by regulating the expression of downstream target gene NF-κB,and itself also could be regulated by NF-κB transcription factors.This essay mainly narrates the expression of microRNA and NF-κB in gastric carcinoma,and summarize the influence to the invasion and metastasis of gastric cancer cells by regulating thier expression interction.
