Clinical significance of Bmi-1 protein expression in colorectal cancer and its precancerous lesion

Objective To analyze the correlation between Bmi-1,Ki-67 and Bcl-2 expression in colorectal cancer and to explore the correlation and significance with clinicopathological features of colorectal cancer.Methods Immunohistochemistry staining was used to display the expression of Bmi-1,Ki-67 and Bcl-2 in 84 cases of colorectal cancer,84 cases of matched normal colorectal mucosa and 21 cases of precancerous lesion.Results The positive rates of Bmi-1 expression in colorectal cancer(73.81%)and intraepithelial neoplasia(95.23%)were significantly higher than those in normal colorectal mucosa membrane(59.52%),P<0.05.There was a positive relationship between Bmi-1 and Bcl-2 expression in 84 cases of colorectal cancer(r=0.358,P<0.001).Conclusion The abnormality of Bmi-1 expression in colorectal cancer might be involved in cell proliferation,apoptosis and cancerization.Bmi-1 was closely correlated with Bcl-2 in colorectal cancer,suggesting that Bmi-1 might coordinate with Bcl-2 to induce anti-apoptosis effect,whereas the suppressing apoptosis pathways were different.
Association of genetic polymorphism in the DNA repair gene XPD and risk of lung cancer in non-smoking females

Objective Xeroderma pigmentosum group D(XPD)is one of the important DNA repair genes.XPD polymorphism at Lys751Gln site and Asp312Asn site have been shown to alter XPD protein function,modulate DNA repair capacity and therefore affect cancer risk.The aim of this study is to explore the relationship between XPD polymorphism and susceptibility to lung cancer in nonsmoking female by a population-based case-control study.Methods There were 222 female patients who were diagnosed with lung cancer from Heilongjiang Tumor Hospital,and the control group included 222 healthy volunteers who were obtained from community centers.XPD genotypes of cases and controls were determined by PCR RFLP method.Unconditional logistic regression analysis was performed to calculate the odds ratios(OR)with 95 confidence intervals(CI)for estimating the association between certain genotypes and lung cancer.Results All of the subjects in this study were nonsmoking females who living in Harbin more than 30 years.The risk of lung cancer was higher in those with the Lys/Gln or Gin/Gin genotype than in those with the Lys/Iys genotype and adjusted OR was 3.36(95 CI:2.29-4.90)and in those with the Asn/Asn or Asp/Asn genotype than in those with the Asp/Asp genotype and adjusted OR was 1.83(95 CI:1.16-2.91).Conclusion The findings indicated that the Lys751Gln and Asp312Asn polymorphisms in XPD gene are associated with the risk of lung cancer in nonsmoking females.

Objective To investigate the expression of PTEN and PIK3CA in pemetrexed-resistant human pancreatic cancer cell line Patu8988,and evaluate their effects on the biological behaviors of pancreatic cancer cell line.Methods The expression of PTEN and PIK3CA in pemetrexed-resistant pancreatic cancer cell line and normal pancreatic cancer cell line were detected by RT-PCR and Western blot analysis,respectively;Transwell test was used to detect the discrepancy between pemetrexed-resistant and non drug-resistant pancreatic cancer cell line.Result Over-expression of PTEN and PIK3CA in pemetrexed-resistant pancreatic cancer cell line Patu8988 were determined both by RT-PCR and Western blot.In pemetrexed-resistant cell line,PTEN and PIK3CA were ascended by 89% and 76% respectively as showed by Western blot,comparing with that of normal pancreatic cancer cell line.Transwell test showed that the capabilities of migration and invasion of pemetrexed-resistant pancreatic cancer cell were significantly weakened than that of normal pancreatic cancer cell line(P<0.05).Conclusion Both PTEN and PIK3CA enhanced abnormally in pemetrexed-resistant pancreatic cancer cell line Patu8988 and the co-existence of high level of PTEN and PIK3CA induced significant decrease in capabilities of migration and invasion of pemetrexed-resistant pancreatic cancer cell,which suggested mechanism of resisting to pemetrexed may involved function of PTEN and PIK3CA.They may also change the biological behavior of cancer cell.

Objective To evaluate the clinical significance of Reg Iα expression in esophageal carcinoma and patients′serum.Methods The Reg Iα expression was investigated by reverse transcription-polymerase chain reaction,immunohistochemistry and ELISA.Results Reg Iα mRNA expressed not only in esophageal cancer but also in esophageal mucosa,but REG Iα mRNA expression was higher in cancer than that in esophageal musosa,P<0.001.Reg Iα protein expression was detected in 21 of 40 cancer(52.5%),no expression in esophageal mucosa.REG I protein expression wasn′t related to the infiltration and size of tumor,P>0.05;but had significant relation with tumor differentiation and lymphatic metastasis,P<0.001.The level of Reg Iα expression in the patients serum with esophageal cancer was significantly higher than that in the normal control group,P=0.011 and its level was significantly decreased after esophageal cancer being resected,P=0.03.Conclusion The expression of Reg Iα related to pathological features of esophageal cancer,the levels of Reg Iα in serum related to the existence of esophageal cancer.

Objective To investigate the differential diagnosis method between brain glioma recurrence and radiation-induced brain injury.Methods 33 patients with glioma recurrence and radiation-induced brain injury were selected,the symptoms and sign data of patients were collected,MRS examination was performed in all patients.Results Glioma recurrence and radiation-induced brain injury patients showed approximately same clinical manifestations,which was no significant difference between two groups;Cho/Cr and Cho/NAA ratios were significantly higher in recurrent glioma than that in radiation injury(P<0.05).Conclusion The results suggest that the MRS examination should be applied to determine glioma recurrence and radiation-induced brain injury.

Objective To establish a method for quantitative detection of CEA mRNA in the peripheral blood and lymph nodes of patients with cdorectal cancer.Methods Real-time RT-PCR was taken to detect the CEA mRNA comparative expression in lymph node,as well as in the peripheral blood.and ECLIA was taken to detect the value of CEA.Results 50 cases of colorectal cancer CEA mRNA positive expression in peripheral blood before surgery was 84%,and the normal control group(10 persons without positive),the difference was statistically significant(P<0.01);CEA-positive rate of 22% compared with the positive expression rate of CEA mRNA difference was statistically significant(P<0.01);CEA mRNA lymph node positive rate 32.3%,compared with the positive rate in peripheral blood CEA mRNA difference was statistically significiant(P<0.01).Conclusions The method of SYBRGREEN real-time PCR is established for the detection of CEA mRNA expression in patients with colorectal cancer.The positive rate of CEA mRNA is relatively higher in peripheral blood of preoperative patients than that in lymph nodes,and is relatively higher than the value of CEA.It is useful for the preoperative diagnosis of patients with colorectal cancer.

Objective To investigate the correlation of PIK3CA gene with differentiation and invasioness of gastric cancer cells.Methods RT-PCR and Transwell invasion chamber were separately used to analyze PIK3CA mRNA expression and invasiveness of three different differentiated gastric cancer cells.Results When the level of PIK3CA expression was higher,the level of cell differentiation was lower and cell invasiveness was stronger.Conclusion PIK3CA mRNA expression level was closely associated with differentiation and invasion of gastric cancer cells in vitro;PIK3CA may play an important role in cell differentiation,invasion and metastasis of gastric cancer,and is a better target for therapy.

Objective To investigate the expression of MGMT and XRCC1 in invasive ductal carcinoma(IDC),and to explore the relationship between clinical factors.Methods Immunohistochemistry(S-P)was performed for MGMT and XRCC1 on 72 specimens of IDC,30 specimens of benign breast lesions.Results The positive expression rate of MGMT in IDC was 70.8%,lower than which expression in benign breast lesions,which had correlation with pathological grades of invasive ductal carcinoma(IDC)and the expression of ER and p53(P<0.05).The positive expression rate of XRCC1 was 52.8%,lower than which expression in benign breast lesions,which had correlation with the pathological grades of IDC.Conclusion They were important pathological signs to evaluate the malignancy degree and prognosis of the breast cancer.These markers play an important role in making sure the treatment prescription for breast cancer patients.

Objective To investigate the efficacy of the Chinese Herbal Medical method of Inviogorating the Spleen,Tonifying the kidney and activating blood to treat chemotherapy-related anemia patients with advanced cancer,who has the problem of recombinant human erythropoietin resistance.Methods A prospective,randomized and control trial was carried out in 70 cases with advanced cancer,These cases were allocated to two groups,Group A(the tial group,n=34)received chinese herbs medicine(the method of Inviogorating the Spleen,Tonifying the Kidney and Activating blood for 8 weeks),Group B(the control group,n=36)received only support treatment。Record the change of haemoglobin,the rate of transfusion,and clinic symptom of anemia.Results The effective rate of treating anemia,Group A was 41.17%,Group B was 20%(χ2=4.29,P=0.023),Group A was better than group B in the decrease the rate of transfusion,14.71% vs 36.11%,and in the improving symptoms.Conclusion The result showed that to chemothery-induced anemia,who has the problem of recombinant human erythropoietin resistance,the method of inviogorating the spleen,Tonifying the kidney and activating blood could increase hemoglobin(Hb),improving symptoms of anemia。It is very safe.It shows the predominance of Chinese Herbal Medical method,and be propitious to patients with advanced cancer,who need chemotherapy.

Objective To investigate the relationship between the immunohistochemistry expression of p53,Ki-67,TopoⅡ,GST,P170 in triple-negative breast cancer(TNBC)and its prognostic significance.Methods The data of 515 in our hospital were retrospectively analyzed.According to the expression of ER、PR、HER-2,we categarized 515 invasive breast cancer into 3 Groups:HR(ER+/PR + HER-2-),triple negative(ER-,PR-,HER2-),and HER+2(ER-,PR-,HER2+).The expression of p53,Ki-67,TopoⅡ,GST,P170 was detected by immunohistochemical technology in 88 cases of TNBC and their prognostic significance were compared with that of the other two subtypes of non-TNBC.The clinicopathological features,disease free survival rate and overall survival rate of these three subtype were analyzed.Results The expression rate of p53,P170,Ki-67,GSTin TNBC(59.5%、78.6%、19.0%、66.7%)is higher than that of non-TNBC.Among 5-year disease free survival in TNBC,the recurrence rate and death rate is 23.9% and 22.7%,and triple-negative breast cancer has an increased likelihood of distant recurrence(hazard ratio,2.46;confidence interval,1.04-5.84;P<0.05)and death(hazard ratio,2.77;confidence interval,1.09-7.02;P<0.05).Conclusions High expression of p53,Ki-67,TopoⅡ,GST,P170 were correlated with the poor prognostic of TNBC.

Objective A retrospective analysis was carried out to investigate the factors of prognostic and recurrence in stageⅢ non-small cell lung cancer.Methods From January 1,1999 to December 31,2004,175 cases with stageⅢ NSCLC received definitive radiotherapy and eligible for the analysis.Establishing 1 year,3-year and 5-year survival rate as endpoints to research the prognosis of NSCLC.Results The investigation was finished at November 9,2009,with a follow-up rate of 95%,those still alive or lost calculated by censoring.The whole group median survival time was 20 months,1,3 and 5 year survival rates were 43%,15% and 3% respectively.Univariate analysis identified 6 significant prognostic clinical factors including T stage,clinical stage,Chemo radiotherapy,radiotherapy interruption,short-term response,acute radiotherapy pneumonia.Cox regression showed that clinical stage,acute radiotherapy pneumonia,short-term response were the independent prognostic factor of patients with stage Ⅲ NSCLC.There is no independent factor associated with local recurrence,clinical stage was an independent factor of distant metastasis.Conclusion The preliminary study results show that the clinical stage of primary tumor,acute radiotherapy pneumonia,short-term response have a significant effect on the survival rate for patients with Ⅲ NSCLC,which provides a reference for the clinical treatment。

Objective To explore the expression of MT1-MMP and VEGF in breast carcinoma tissue and their association with the clinicopathological features of breast cancer.Methods The immunohistochemical staining method was used to evaluate the expression of MT1-MMP and VEGF in 67 specimens of breast carcinoma and adjacent normal tissues,and its correlations to clinicopathological features were analyzed.Results The expression level of the MT1-MMP and VEGF in breast carcinoma tissues was markedly higher than that in the paraneoplastic breast tissue(P<0.05).The expression of MT1-MMP was positively correlated with the expression of VEGF in breast carcinoma tissues(P=0.005).The expressions of MT1-MMP and VEGF were positively correlated to TNM stage and axillary lymph node metastasis(P<0.05).Conclusions The increasing expressions of MT1-MMP and VEGF were correlated with invasion and metastasis of breast carcinoma,combined examination of VEGF and MT1-MMP can be of value in assessment of the malignancy and in prognostic evaluation of breast carcinoma.
Combined therapy with Kanglaite injection and intraperitoneal catheter drainage with drug to treat malignant ascites

Objective To evaluate the efficacy and safety of intervenous drop infusion of Kanglaite injection and intraperitoneal catheter drainage combined with hydroxycamptothecin and bruceolic oil emulsion in advanced tumor patients with malignant ascites.Methods 65 advanced tumor patients with malignant ascites were chosen and divided randomly into two groups:treatment group and control group.37 patients of treatment group were given hydroxycamptothecin 20 mg,bruceolic oil emulsion 60 ml by intraperitoneal injection with a repetition every week,4 times altogether.Simultaneously,an intervenous drop infusion of Kanglaite injection 200 ml was given every day,21 day altogether.And 28 patients of control group were given DDP 80-100mg by intraperitoneal injection with a repetition every week,4 times altogether.The effects were evaluated after a month.Results The malignant ascites reduced:the treatment group efficient 78.38%(29/37),and the control group efficient 50%(14/28).The toxic reactions and adverse reactions of treatment group were less than that of control group.The patients of treatment group received a better quality of life.Conclusion Intraperitoneal catheter combined with hydroxycamptothecin,bruceolic oil emulsion and intervenous drop infusion of Kanglaite injection is feasible and effective for advanced tumor patients with malignant ascites.
