Associations between CYP24A1 gene polymorphisms and the risk and prognosis of colorectal cancer

Objective To explore the associations between polymorphisms in CYP24A1 gene and the risk and prognosis of colorectal cancer (CRC).Methods A case-control study consisting of 528 CRC patients and 605 cancer free controls and a follow up study with 317 cases were conducted to explore the associations between two polymorphisms in CYP24A1 and the risk and prognosis of CRC,as well as the combinations and interactions of polymorphisms with dietary factors on CRC risk.Results Although there was no association between polymorphisms in CYP24A1 and the risk of CRC,significant combinative effects between polymorphisms and dietary factors on CRC risk were observed.For Rs4809957 polymorphism,the prognosis of AA genotype carriers was worse than GG/GA carriers in CRC,colon cancer and rectal cancer(Log-rank test P=0.01,P=0.01,P=0.02,respectively).Compared with GG genotype,AA genotype carriers of Rs4809957 polymorphism had worse prognosis in CRC(HR = 2.35,95% CI = 1.28~4.30),colon caner(HR = 2.37,95% CI =1.09~5.14)and rectal cancer(HR = 2.11,95% CI = 1.11~4.01).Conclusion The combinations of the polymorphisms in CYP24A1 gene with dietary factors are associated with the susceptibility of CRC,and Rs4809957 polymorphism may lead to a worse prognosis of CRC.

Objective To investigate the relationship between HIF-1α and bevacizumab resistant in ovary cancer,and explore the influence on ovary cancer cell by HIF-1α.Methods The correlation was analyzed between HIF-1α expression and clinicopathological parameters using immunohistochemistry in 62 patients with ovarian cancer.In addition,the expression of HIF-1α and VEGF were evaluated by real time PCR or Western Blot in SKOV3 and ES-2 cell under CoCl 2 treatment.Results HIF-1α was overexpressed in cancer tissue,and its high levels were related to CA125(P=0.027)and resistant to bevacizumab(P<0.001).Moreover,CoCl2 could induce high expressions of HIF-1α and VEGF,when compared to normal condition.Conclusion Our findings suggest that HIF-1α is associated with resistant to bevacizumab.It is more chemotherapeutic sensitivity when combined with bevacizumab and HIF-1α antibody.

Objective To identify and analyze the DNA methylation induced pathogenic genes in breast cancer.Methods Using genomic data provided by the international cancer genome,we carried out systematic analysis of the abnormal gene expression in breast cancer and epigenetic regulation mechanism by bioinformatics Methods .Results We identified 428 genes with abnormal expressions in breast cancer by t test.Functional enrichment analysis revealed genes up-regulation in breast cancer were closely related to the cell cycle,and those down-regulated were significantly enriched in the hormone response function.Study on DNA methylation revealed that breast cancer showed an unique DNA methylation pattern.Further analysis reveals 23 breast cancer genes induced by abnormal DNA methylation.Conclusion DNA methylation can mediate the abnormal expression of breast cancer genes,and is an important biological marker for early diagnosis and prognosis of breast cancer.

Objective To analyze the clincopathological features and prognosis of 77 colorectal cancer patients with synchronous liver metastases after resection of the primary tumors.Methods Seventy seven colorectal cancer patients with synchronous liver metastases who underwent resection of the primary tumors and pathological confirmed were enrolled in this retrospective study.The clincopathological features and prognosis were analyzed.Kaplan-Meier method was used to analyze survival rate.The differences between survival curves were calculated by Log-rank test.Results The analysis showed that sex,tumor differentiation and TNM stage were associated with prognosis(P<0.05).The analyses demonstrated no differences in age,primary tumor site,treatment,number of liver metastases and extrahepatic metastasis(P>0.05).There was a significant correlation between CEA,CA19-9 level and gender,tumor differentiation and TNM stage(P<0.001)and the corresponding surgical procedure and TNM stage(P<0.05).Conclusion The analysis show that male,the low differentiation and stage Ⅲ are poor prognosis factors.

Objective To compare the prognosis efficacy between total laparoscopic gastrectomy and ordinary gastric cancer for familial gastric cancer.Methods The selected 60 patients with ordinary gastric cancer from August 2012 to 2014 in our hospital as the control group,the same period in familial gastric cancer of 40 patients were chosen as control group.All patients were performed by total laparoscopic gastrectomy.Results The operative time,blood loss,postoperative discharge time compared in the two groups were showed no significant differences.However,the postoperative hospital stayed in the observation group and the number of lymph nodes were significantly longer more than the control group(P<0.001).The postoperative pulmonary infection,intestinal obstruction,abdominal infection,anastomotic leakage,anastomotic bleeding and other complications in the observation group were significantly lower than in the control group(P<0.001).After the investigation,the dysphagia,pain,anxiety,physical appearance,dry mouth scores in the observation group were significantly higher(P<0.001).Conclusion Familial gastric cancer has unique clinical and pathological features,Compared to ordinary stomach,the total laparoscopic gastrectomy treatment of familial gastric cancer need more lymph nodes dissection,it can cause more complications,it has some impacts on the quality of life for patients.

Objective To evaluate Results of cancer early detection and early treated project screening and experience in Heilongjiang province from 2013 to 2014.Methods In Harbin and Daqing city resident population,high-risk groups were screened through the risk factors questionnaire and screened corresponding clinical examine,respectively(lung cancer,liver cancer,breast cancer,upper gastrointestinal cancer and colorectal cancer).To explore cancer preventive effect of early detection and early treated in Heilongjiang province.Results We smoothly completed the cancer early detection and early treatment between 2013—2014 in Heilongjiang province,a total of 15628 people received a copy of risk factors questionnaire,10299 people in high-risk groups received clinical screening.A total of 66 cases of suspected cancer cases were screened.Conclusion completed Cancer early detection and early treatment are performed in Heilongjiang province between 2013—2014.Early detection can be treated early project for early discovery,early diagnosis and early treatment.It is important to improve the quality of screening survival time and survival of patients′.The project also promotes the early diagrosis and early treatment experience in Heilongjiang province.

Objective Vimentin is the major protein consisting of intermediate filaments in normal and neoplastic mesenchymal cells.It has been regarded as a marker of epithelial mesenchymal transition(EMT).However,little is known about colorectal cancer(CRC)with Vimentin expression as a marker of EMT and prognosis.Methods We investigated the expression of Vimentin in CRC tissues and determined the correlations between Vimentin expression patterns and clinicopathologic characteristics and prognosis in CRC.In total,189 patients diagnosed with CRC based on histopathological evaluation and those subjected to surgical resection at the Liaoning Province Cancer Hospital and Institute between 2000 and 2002 were examined.Immunohistochemical staining for Vimentin was performed for each specimen.Results Positive expression of Vimentin was significantly associated with the lymph node metastasis(pN stage,P=0.038),TNM stage(P=0.024).Survival analysis revealed that positive Vimentin expression was significantly related to poor prognosis(P=0.016).Multivariate analysis revealed that Vimentin expression was an independent prognostic factor for patient survival in colorectal cancer(P=0.026).Conclusion In Conclusion ,our data provide novel evidence for the clinical significance of Vimentin expression as potential predictive biomarkers for identifying patients with lymph node metastasis or poor prognosis in CRC.

Objective To improve the clinical diagnosis and prognosis of the treatment of primary liver carcinoma with bone metastasis.Methods A retrospective study on diagnosis and treatment of bone metastasis from 55 cases of primary liver carcinoma was developed.Survival rates were calculated by Kaplan-meier method,univariates analysed by Log-rank and multivariates analysed by Cox regression.Results The 1-,2-and 3-year cumulative survival rates of the cases from primary liver carcinoma were 54.5%,25.5% and 16.4% respectively.The mediate survival time was 13 months.And the cases with bone metastasis were 23.6%,10.9% and 1.8% respectively.The mediate survival time was 5.5 months.Factors such as metastasis to other organs,liver function,and the combined modality therapy were independent prognostic factors.While number of bone metastasis,AFP level and number of liver cancer had no significant relations with the survival rate.Conclusion The prognosis of bone metastasis from primary liver carcinoma is poor.It is important to take emphasis on combined modality therapy,which may be benefit on reducing the symptom,improving the quality and prolong the life span.

Objective To evaluate the effect and mechanism of ginseng polysaccharides injective solution on invasive ability of human breast cancer MCF-7 cells.Methods Human breast cancer MCF-7 cells were treated with different concentrations of ginseng polysaccharides injections(12.5μL/mL,25μL/mL,50μL/mL or 100μL/mL),and phosphate buffer saline was served as control.The invasive ability,transfer ability and protein expression of matrix metalloproteinases-2(MMP-2)and matrix metalloproteinases-9(MMP-9)were determined via transwell matrigel invasive test,scratch test and Western Blot,respectively.Results Ginseng polysaccharide injection on invasive ability and transfer ability in vitro of MCF-7 cell had significant role in inhibiting effect,and showed dose and time dependent.After ginseng polysaccharides injection treatment,the MMP-2 and MMP-9 protein expression significantly reduced,and showed dose and time dependent manner.Conclusion Ginseng polysaccharides injection is able to inhibit the invasion and transfer in MCF-7 cells.Moreover,the regulation of MMP-2 and MMP-9 expression may be involved in this effect.

Objective To investigate the expression of AICARFT and mTOR in lung adenocarcinoma cell line A549 and pemetrexed resistant human lung adenocarcinoma cell line A549/PEM.Methods The distribution of cell cycle was evaluated by flow cytometry assay.The expression of AICARFT and mTOR in A549 and A549/PEM were detected by RT-PCR and Western blot.Results Flow cytometry analysis showed that S phase in A549/PEM was higher than that in A549((36.61±1.36)% vs(27.16±1.08)%(P=0.001)).mRNA over expression of AICARFT and mTOR in A549/PEM were detected by RT-PCR(P<0.05).Protein over expression of AICARFT and mTOR in A549/PEM were detected by Western blot.(P<0.05).Conclusion High level of AICARFT and mTOR may be connected with pemetrexed acquired drug resistance in lung adenocarcinoma.The cycle of pemetrexed resistant human lung adenocarcinoma cancer cell line A549/PEM is redistributed.

Objective To evaluate the clinical effect of cytokine induced killer cellular immunotherapies(CIK)on the patients with advanced lung cancer.Methods Peripheral blood mononuclear cell from 32 cases of patients with advanced lung cancer were isolated.Cells were cultured in DMEM supplemented with IFN-γ,IL-2,anti-CD3 monoclonal antibody and other cytokines,and transfused back into the patients in several times after 15 days of culture.Subsequently,we observed the size of tumor,the change of immunologic function by CIK treatment,the status of clinical symptoms,and the toxic and side effect.The clinical curative effect was also evaluated.Results The symptoms in 32 patients who received CIK therapy were reduced,and the side effects were light.Immunophenotype analysis showed that CD3 / CD4 coexpression were found in 34.4% patients,CD3 / CD8 coexpression in 68.2%,and CD3 / CD56 co-expression in 13.7% patients.CT images about 15 days after CIK cell transfusion showed that pleural effusion were decreased.Clinical evaluation effective rate was 56.0%,and disease control rate(CR + PR + SD)was 84.3%.Conclusion CIK cell immunotherapies of cancer patients can improve immunologic function of cancer patients and inhibit tumor growth and relieve the clinical symptom of patients,and improve the quality of life.

Esophageal cancer with eyeball metastasis is scarce,and the clinical diagnosis is based on ultrasound.The treatment method is surgical removal at present.Our focus is through other local or systemic treatment to protect eyeball,protect the integrity intact and facial appearance.One patient&apos|s clinical data with esophageal eyeball metastasis from the People′s Liberation Army Cancer Center of No.105 Hospital was included.The patient was completed “enucleation of the right side of eyeball” by ultrasound diagnosis.The Results of pathological suggested that poorly differentiated squamous cell carcinoma metastasis in the eye.Eyeball metastases′s operation was received a good recovery after 2 months and there was no recurrence.In the present paper,we discuss esophageal eyeball metastases clinical characteristics and Methods of diagnosis and treatment combined with the literature.

Tumor microvessel including two modes,as one is classic vascular structures surrounded with vascular endothelial cells and the other is vasculogenic mimicry(VM)surrounded with cancer cells.Recent studies suggested that anti-angiogenic drugs can inhibit classic microvessel structure significantly,but can not inhibit VM formation and even promote VM formation further more.This is one of the reasons affecting its clinical efficacy.However,basic research has shown that traditional chinese medicine,gene therapy and drugs which is targeting for VM formation have an unique advantage in anti-VM.In this review,we summarize the progress of cancer therapy on targeting vasculogenic mimicry,and provide a basic for the development of new drugs for cancer therapy through targeting both angiogenesis and VM formation.

Breast cancer is one of the most common malignant tumors in women.Early diagnosis plays a very important role in the treatment of breast cancer.Recently,with the development of epigenetics,it gives us a new orientation for early detection for cancers.The most understood mechanism of epigenetics is DNA methylation.Ras associated domain family 1A gene(RASSFlA)promoter methylation occurres in tumor′s early stage,and have a statistically concern with tumor′s TNM stage,invasiveness and outcomes.Moreover,DNA methylation can be reversed,so RASSF1A can be a promising candidate for early breast cancer detection biomarker,and can be a potential orientation for breast cancer treatment.

With development of molecular biology,breast cancer has entered an era of molecular classification,thus making biomarker based personalized medicine is the trend of breast cancer treatment.HER2-positive breast cancer of high invasiveness and bad prognosis accounts for 20%~30%.Targeting HER2,trastuzumab is the first humanized monoclonal antibody which can improve the prognosis of HER2-positive patients and it is recommended by guidelines and expert consensus at home and abroad for anti-HER2 therapy in any stage.However,the cardiotoxicity,de novo resistance and acquired resistance of trastuzumab make the clinician to explore the second line anti-HER2 therapy.Lapatinib is the first FDA approved and HER1,HER2 double-targeting tyrosine kinase inhibitor which can be a better choice after failure with trastuzumab.This article reviews the application,some clinical and mechanism of drug resistance researches of Lapatinib.

Testicular proteoglycan 1(SPOCK1),a kind of extracellular matrix glycoprotein,can inhibit the activity of cathepsin and promotes the combination of low affinity for calcium.SPOCK1 is essential to the development of mammals.It not only regulates cell,and the interactions between cells and matrix are also related to cell migration and proliferation.Currently,SPOCK1is confirmed that expression can promote the occurrence and development of tumor,suggesting it could be a new anti-tumor target.This article is to review SPOCK1 molecular structure,biological function and its relationship with tumor.
